This kit provides a method for biotinylating DNA probes through incorporation of biotin-dCTP via a thermostable DNA polymerase and specific primers in the polymerase chain reaction (PCR). Large quantities of biotinylated probes can be generated from a small amount of template. A template ranging in size from 100 bp to 1.1 kbp is required. Reaction requires only 10 pg (plasmid) or 1 ng (genomic) template DNA per reaction. DNA probes are stable for up to 1 year without loss of activity.
Quantitation standard allows for determination of probe specific activity. Biotinylated PCR probes can be detected using enzyme-labeled streptavidin and a chemiluminescent or chromogenic substrate in southern blots, northern blots, dot blots, in situ hybridization, colony lifts, and plaque hybridizations.
